Evaluation of Biochemical Markers in Hepatocyte like Cells Differentiated from Adipose Derivation Stem Cells

چکیده
Recent evidence proposes that cells with the properties of mesenchymal stem cells (MSCs) can be extracted from adult peripheral tissues, including muscle, dermis and adipose tissue. Adipose tissue is ubiquitous and easily obtainable in large quantities. We separated Adipose derived stem cells (ADSCs) from part of subcutaneous adipose tissue from rat. These cells could be readily differentiated into cells of the chondrocyte, osteocyte, hepatocyte and adipocyte lineage demonstrating their multipotency. It is necessary to describe the ADSCs surface expression of the typical markers by using flow cytometric and survey the results. Finally, these cells transformed into hepatocyte like cell. Immunocytochemical analysis demonstrated that ADSCs are capable to express Albumin and -fetoprotein during differentiation. Moreover, we determined biochemical markers of hepatocyte like cells such as SGPT, SGOT, urea Production and glycogen synthesis. At last we isolated total RNA from hepatocyte like cells at shown days after differentiation and performed RTPCR by using the specific primers (albumin, Fetoprotein and Alpha 1-Antitrypsin).

Development of Simple Protocol for Generation of Functionally Active Hepatocyte-like Cells from Human Adipose Tissue-derived Stem Cells
Background and Aims: Human adipose tissue-derived stem cells (hASCs) are considered as an attractive source of regenerative stem cells, mainly because of their higher proliferation rate, more accessibility and hepatocyte like properties as compared to mesenchymal stem cells isolated from other tissues. Numerous studies have described the beneficial use of adipose tissue-derived stem cells for g...

Objective(s) Bone marrow is the traditional source of human multipotent mesenchymal stem cells (MSCs), but adipose tissue appears to be an alternative and more readily available source. In this study, rat adipose-derived stem cells (ADSCs) were induced to differentiate into Schwann-like cells and compared with rat bone marrow stem cells (BMSCs) for their Schwann-like cells differentiation pote...

Background: Wharton’s jelly is an unlimited source of stem cells that can be used in cell therapy and tissue engineering without any ethical concern. It has been revealed the cell-free extract could be effective to induce cell differentiation. The objective of this study was to induce Wharton’s jelly-derived mesenchymal stem cells (MSCs) into hepatocyte-like cells by premeabilization of the cel...

Background: Adult stem cells (ASC) are undifferentiated cells found throughout the body. These cells are promising tools for cell replacement therapy in neurodegenerative disease. Adipose tissue is the most abundant and accessible source of ASC. This study was conducted to evaluate effect of selegiline on differentiation of adipose-derived stem cells (ADSC) into functional neuron-like cells (NL...
